I have one drive 500GB. I am using acronis to have backups over there.
IS there any way to hide that drive from explorer or make that drive only used by acronis and other read only
Just like what windows backup does.

Or if you are using a Professional or higher level of Vista or 7 then you can configure a group policy to hide the drive... .
See these instructions on how to hide a drive from Explorer .
If you are running Windows XP, then using Tweak UI is your best option .

Do I have to create a full backup plan before a incremental back up plan?

If I create an image backup (a la Acronis True Image or Ghost, etc) of the VM host's system drive and then restore that image, will my VM's still work as if nothing ...

You may have to re-configure some areas however if things change... .
The VM software and the VM's are part of the host system .
If you are taking an image of the entire host, then when it is restored, it should be in exactly the same state, including the VM's .
