I am planning to go to Pisa from Rome by a regional train. Is there a direct train ? or I have to change in Florence ? In case I have to change can I purchase the ticket for the whole journey in Rome itself ?

So, we're 99% sure we will start our Italian adventure in Rome this May. If you've been in the past few years, where did you stay?
What do you suggest as a "can't miss" type thing?
Thanks, peeps! :-)
