What's the best way to find out if a user is not using IE 7 or 8, Firefox, Safari, Chrome or Opera? I want to show a message if they are not using one those browsers. In this case is it just easier to use browser sniffing?

The problem is, in the future when a new browser is released and becomes more modern than those with older browsers that they need to upgrade, then pick some features that your site needs in order locked into hard coding a list of compliant....
Do all browsers support PHP's $_SERVER['HTTP_X_REQUESTED_WITH']? Do you know of any browsers or times where $_SERVER['HTTP_X_REQUESTED_WITH'] would not work?

Sending of a X-Requested-With header really depends.
Do all browsers support setting custom request headers for XMLHttpRequest? As far as I know, yes, it works in all modern browsers.

Place favicon.ico at ....
Modern browsers also support PNG browsers.
You can; Favicon must be an .ico file to work properly on all browsers.
Having a favicon.* in your root directory is automatically detected by most browsers.
