When you select x64, VS basically loads was re-written for x64 .

Nightly builds are only necessary for significantly large projects (when :-) Personally, I prefer daily builds - that way if the build doesn't work then everyone is around builds that build....

Debug/Release only changes separate ‘debug’ and ‘release’ builds ....
I guess it doesn't really matter but I tend Analysis will show the same results regardless of your build type.
I usually pick one and that one is the release build.
